Research Services Core Facility

Pathology Research Services Core Facility provides UC Irvine Health researchers with dependable and economical assistance in obtaining and analyzing human tissue, blood and serum samples. The core facility leverages the existing professional expertise and technical resources available in the facility Department of Pathology & Laboratory Medicine's various clinical laboratories.

Compliance

Since core services require the use of tissue derived from either humans or vertebrate animals in almost all cases, investigators are required to document that research projects have been subjected to either the Institutional Animal Care and Use Committee (IACUC) or the Institutional Review Board (IRB) when requesting services. This documentation includes a copy of the approval letter, which is kept on file. Compliance with these institutional policies is rigorously enforced. Failure to supply the necessary information will result in a delay in the provision of requested services.

The IRB revised the rules governing human subject research on de-identified specimens in November 2006. Thus, use of tissues from our banks in this manner no longer requires explicit IRB approval. To ensure investigators understand the specific limitations of the use of de-identified tissues, we ask that they file a Request for Determination of Non-Human Subject Research (PDF). A copy of the same form must be faxed or emailed to the appropriate departmental personnel dispersing tissue, in lieu of IRB approval.

Cost to Investigator

The activities of the core are underwritten in part by funds from the National Institutes of Health (NIH) in the form of a Cancer Center Support Grant and by the Department of Pathology & Laboratory Medicine. Investigators are recharged for the marginal costs associated with each service. For some services, a specific order form with recharge rates is determined for individual projects, in consultation with the appropriate associate director.

The research core consists of five component services:

Experimental tissue resource (histology and Immunohistochemistry) »

Services Offered

Services provided by this component of the core include routine histology (tissue trimming, embedding, sectioning, including frozen sections, H&E staining, histochemical staining and immunohistochemical stanining), and the retrieval of paraffin-embedded blocks of normal and diseased tissue (by diagnosis) from our archives. A detailed list is provided on the order form.

IRB approval is required for the use of human tissue. IACUC approval is required for the use of animal tissue. Service requests will not be processed without the necessary compliance information.

Ordering Services

Download the order form. Fax the completed form to 714-456-5873 or email it to Jeff Kim in histology at jbkim@uci.edu.

Investigators may use our departmental courier service to send samples to UC Irvine Medical Center, where the histology unit is located. Processed samples can also be returned by courier.

To send samples via courier, email the request form to Jeff Kim (specifying that samples will follow via courier) and include a copy of the request form with the samples. Pack the samples securely in a sturdy box. Tightly seal any containers filled with formalin. Be sure to specify that samples are to be returned by courier also. Take the box to the courier pickup location in the Med Sci D476 between 10 a.m. and 6 p.m. on the UC Irvine School of Medicine campus.

Morphometry and image analysis of tissue sections »

Services Offered

This component of the core provides services required to quantify cellular elements or molecular signals detectable by microscopic evaluation of tissue sections. This includes determining the number and/or size of cellular organelles; measuring the intensity of immunostaining of individual cells in a tissue section, and counting the percentage of cells and/or nuclei that are immunostained with a particular antibody. These analyses can be performed on sections which were H&E stained or immunostained in the histology component of the core, or on sections prepared by individual investigators.

Cost to Investigator

The recharge rate for this service is $18 per hour of data acquisition.

Laser-capture micro-dissection of tissue sections »

Services Offered

This component of the core allows investigators to fractionate cell populations from tissue sections. For example, normal tissue can be purified from tumor tissue on tissue sections containing a mixture of these cell types. The core facility contains a Leica CM3050 cryostat and a Leica LS-AMD outfitted with fluorescent optics (allowing the selective visualization and capture of immunostained cell populations, if so desired). We also provide access to an Arcturus XT, with UV and IR laser capture modes, to facilitate the capture of single cells and live cells with minimal damage to biomacromolecules. The instrument is located in the CLRB (Building 54, 4th floor) at UC Irvine Medical Center.

This service can be performed on sections that were H&E stained in the histology component of the core or on sections prepared by individual investigators.

Cost to Investigator

All users must first undergo a two-hour orientation on the instruments with Dr. Robert Edwards, assistant professor of pathology. There is no fee for UC Irvine investigators, and $100 per hour for non-UC Irvine affiliated users. All investigators requiring the services of a surgical pathologist to perform the microdissection will be charged $150 per hour of LCM time. Investigators must purchase appropriate caps with laser-capture membrane (Arcturus) or foiled-coated slides (Leica).

Tissue biorepository »

Services Offered

The core will bank remnants of tissue (both normal and tumor) resected from patients undergoing surgery at UC Irvine Medical Center. Remnant tissue is the residual portion of a surgical specimen following processing for diagnostic purposes (see Tissue Custody Policy). Signed consent from each patient will also allow us to obtain peripheral blood samples (serum and DNA) and clinical follow up at six-month intervals for three years. Investigators may request access to tissue and blood samples, as well as clinical data. Requests for large amounts of tissue or multiple samples, or access to relatively rare specimens will be reviewed by the Core’s Tissue Use Committee.

IRB approval is required for access to this tissue. Service requests will NOT be processed without the necessary compliance information. Investigators are encouraged to contact Dr. Dan Mercola concerning the availability of tissue prior to submitting projects for IRB approval.

Genotyping (includes nucleic acid extraction from blood and tissue sections) »

Services Offered

Genotyping services are performed in the Diagnostic Molecular Pathology lab and include a variety of PCR and/or Southern blot-based analyses of mutant or polymorphic alleles. The lab will work with investigators to identify genotype-based tests that may be of value in clinical treatment and cancer control studies. This service can also perform mRNA-based testing for occult cancer or residual disease. In addition, this component of the Core provides automated nucleic acid extraction from whole blood, paraffin-embedded or frozen tissue sections.

Nucleic acid extraction recharge rates are listed on the order form. Recharge rates are determined on a case-by-case basis.
